Detection of ICAM-3/CD50 in Human Blood Lymphocytes by Flow Cytometry. Human peripheral blood lymphocytes were stained with (A) Mouse Anti-Human ICAM-3/CD50 Monoclonal Antibody (Catalog # MAB8131) or (B) Mouse IgG1 Isotype Control (MAB002), followed by Allophycocyanin-conjugated anti-Mouse IgG secondary antibody (F0101B) and Mouse Anti-Human CD3 epsilon PE-conjugated Monoclonal Antibody (FAB100P). Staining was performed using our Staining Membrane-associated Proteins protocol.
ICAM‑3/CD50 in Human Lymph Node. ICAM‑3/CD50 was detected in immersion fixed paraffin-embedded sections of human lymph node using Mouse Anti-Human ICAM‑3/CD50 Monoclonal Antibody (Catalog # MAB8131) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell surface of lymphocytes. Staining was performed using our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Background: ICAM-3/CD50
Intercellular Adhesion Molecule-3 (ICAM-3; also known as CD50), a member of the immunoglobulin superfamily, binds the leukocyte integrins LFA-1 (CD11a/CD18) and alpha d/ beta 2. ICAM‑3 is expressed on leukocytes and epidermal Langerhans cells. It may play an important role in T cell stimulation by Langerhans cells. ICAM-3 has not been identified in the mouse.
