UM-UC3 bladder cancer cells were transiently transfected with a Notch-reporter Firefly luciferase plasmid, in addition to an internal Renilla control. IgG was used as a negative stimulation control, whereas Jagged 1 was used to induce a Notch-specific response. Increasing concentrations of the Gamma-secretase inhibitor DAPT resulted in a dose-dependent decrease in Notch signal.

Used at 10 ug/mL works perfectly as a human Isotype control for flow cytometry. There is no background binding in live cells that do not express Fc receptor. The attached image shows MFI with and without the isotype control indicating identical signal.
