Immunology News: Targeting Myeloid-Derived Suppressor Cells (MDSCs) for Cancer Immunotherapy

星期二, 七月 3, 2018 - 08:24
MDSC

Myeloid-derived suppressor cells (MDSCs) are a diverse population of immature myeloid cells with immunosuppressive properties that accumulate under pathological conditions including specific types of cancer and infections. Two primary subsets of human and mouse MDSCs known as granulocytic and monocytic MDSCs have been identified. These cells are of great interest as high levels of circulating MDSCs in cancer patients have been shown to correlate with clinical stage, metastatic burden, and resistance to both chemotherapy and immunotherapy. As a result, MDSCs are now recognized as a major obstacle in the treatment of cancer. Multiple strategies targeting these cells are being investigated to determine if the...

Validating Antibodies to Support the AstraZeneca Drug Discovery Pipeline

Validating Antibodies to Support the AstraZeneca Drug Discovery Pipeline

Antibodies are critical tools in the majority of research and clinical studies. However, unreliable antibodies with poor specificity or reproducibility remain a significant drain on resources across biomedical research and industry. Widespread acceptance of the need for end-user antibody validation has led to high profile initiatives and guidance (e.g. Uhlen et al, Nat. Methods 2016). Aligning to these principles, the AstraZeneca Affinity Reagent Group provides company-wide advice and scientific support in the validation of both commercial and our bespoke antibodies generated in-house. Our work feeds into activities across the drug discovery pipeline, including target identification / validation, hit-...

The Heat Is On: Detecting Chemokine Receptors on Live Cells

星期五, 六月 15, 2018 - 13:14
CCR7

Chemokine receptors and other multipass proteins require special consideration when staining live cells for flow cytometry analysis. These receptors have seven transmembrane domains, all of which are fluid and moving around in the cell membrane. As a result, the three extracellular loops, to which antibodies must bind, are also moving and this motion can change the orientation of antibody binding sites. While this domain flexibility is necessary for ligand binding to the receptor, it can create problems for flow cytometry staining of live cells. In addition to movement within the lipid membrane, chemokine receptors can undergo internalization and endocytic recycling,...

What Are the AT(N) Biomarkers for Alzheimer’s Disease?

星期五, 六月 8, 2018 - 10:42
Atlanta Biological Fetal Bovine Serum and Media

Researchers have long been attempting to identify and validate specific biomarkers for Alzheimer’s disease (AD). AD is a neurodegenerative disorder that is characterized by brain atrophy resulting from neuronal loss. It is typically diagnosed following a series of clinical assessments that include neurological and mental examinations and brain imaging. However, since many other disorders also display AD-like symptoms, the only way to definitively diagnosis AD is by tying clinical assessment of the disease to the postmortem identification of its pathological hallmarks (i.e. senile plaques...
