Why Use Cultrex 3-D Spheroid Basememnt Membrane Extract Cell Invasion Assay, 96-well?

The Cultrex 3-D Culture Spheroid Cell Invasion Assay utilizes a 3-D Culture Qualified 96 Well Spheroid Formation Plate alongside a specialized Spheroid Formation Extracellular Matrix to drive aggregation and/or spheroid formation of cells. Upon completion of spheroid formation, the spheroid is embedded in an invasion matrix composed of basement membrane proteins. This matrix forms a hydrogel network on which invasive cells can travel. Invasion modulating agents can be applied to the system to evaluate the impact on cell response. Cell invasion is visualized microscopically and can be quantified through image analysis software.

Embedding Spheroids in Cultrex BME for Cell Invasion and Migration Assays. MDA-MB-231 (3,000 cells/well) formed spheroids in low adhesion plates for 72 hours using reagents in the Cultrex 3-D Spheroid Basement Membrane Extract Cell Invasion Assay. Briefly MDA-MB-231 cells were cultured in the presence of 1X Spheroid Formation Matrix. Then spheroids were embedded in A) no Invasion Matrix, or B) Spheroid Invasion Matrix. After hydrogel polymerization, DMEM and 10% FBS was added to each well to promote a chemotactic response over a 96 hour period. Note the addition of the Spheroid Invasion Matrix enhanced cell invasion over spheroids cultured without embedding in the invasion matrix.
