Detection of CD8 alpha in Human Blood Lymphocytes by Flow Cytometry. Human peripheral blood lymphocytes were stained with (A) Mouse Anti-Human CD8 alpha Monoclonal Antibody (Catalog # MAB1509) or (B) Mouse IgG2B isotype control antibody (Catalog # MAB0041) followed by anti-Mouse IgG PE-conjugated secondary antibody (Catalog # F0102B) and Mouse Anti-Human CD3 epsilon APC-conjugated Monoclonal Antibody (Catalog # FAB100A). CD8 alpha in Human PBMCs. CD8a was detected in immersion fixed human peripheral blood mononuclear cells using Mouse Anti-Human CD8a Monoclonal Antibody (Catalog # MAB1509) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 493-conjugated Anti-Mouse IgG Secondary Antibody (green; Catalog # NL009) and counterstained with DAPI (blue). Specific staining was localized to cell surfaces. View our protocol for Fluorescent ICC Staining of Non-adherent Cells.

Background: CD8
CD8 is a heterodimeric glycoprotein consisting of an alpha and beta chain. It is expressed on cytolytic T cells and functions in conjunction with the T cell receptor in the recognition of MHC/peptide complexes.
