Detection of Human LEDGF by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line. Gels were loaded with 40 µg of cytoplasmic (Cyto) and 20 µg of nuclear (Nuc) extracts. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human LEDGF Monoclonal Antibody (Catalog # MAB3468) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). Specific bands were detected for LEDGF p75 at approximately 75 kDa and LEDGF p52 at approximately 50-55 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

LEDGF in HeLa Human Cell Line. LEDGF was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human LEDGF Monoclonal Antibody (Catalog # MAB3468) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red, upper panel; Catalog # NL007) and counterstained with DAPI (blue, lower panel). Background: LEDGF
LEDGF, also known as transcription co-activator p75, is a 75 kDa protein that belongs to the hepatoma-derived growth factor (HDGF) family of nuclear targeted proteins. It is a widely expressed protein that interacts with lentiviral integrase. As a result of alternative splicing, isoforms with deletions, substitutions/insertions have been reported. Human LEDGF shares 91%, 92% and 96% amino acid sequence homology with rat, mouse and feline LEDGF, respectively.
