Detection of Human Phospho-PLC‑ gamma 1 (Y783) by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line untreated (-) or treated (+) with 1 mM Pervanadate (PV) for 5 minutes. PVDF membrane was probed with 0.05 µg/mL of Rabbit Anti-Human/Mouse/Rat Phospho-PLC- gamma1 (Y783) Monoclonal Antibody (Catalog # MAB74541) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-PLC- gamma1 (Y783) at approximately 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: PLC-gamma 1

Phospholipase C gamma-1 (PLC-gamma 1) is activated by receptor tyrosine kinases in response to growth factors and hormones, and phosphorylation at Tyr783 is a widely used indicator of activation. Activated PLC-gamma 1 catalyzes the hydrolysis of phosphatidylinositol 4,5-biphosphate to produce the second messengers inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG). IP3 mobilizes the release of calcium while DAG activates protein kinase C. PLC-gamma 1 plays an important role in regulating cell proliferation and differentiation.
