Detection of Human eNOS by Western Blot. Western blot shows lysates of HUVEC human umbilical vein endothelial cells untreated (-) or treated (+) with 100 µM Pervanadate (PV) for 10 minutes. PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human Phospho-eNOS (S1177) Monoclonal Antibody (Catalog # MAB9028) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for eNOS at approximately 140 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human eNOS by Simple WesternTM. Simple Western lane view shows lysates of HUVEC human umbilical vein endothelial cells untreated (-) or treated (+) with Pervanadate (PV), loaded at 0.2 mg/mL. A specific band was detected for eNOS at approximately 141 kDa (as indicated) using 10 µg/mL of Rabbit Anti-Human Phospho-eNOS (S1177) Monoclonal Antibody (Catalog # MAB9028). This experiment was conducted under reducing conditions and using the 66-440 kDa separation system.

Background: eNOS
Endothelial NOS (eNOS), also known as nitric oxide synthase 3 (NOS3) or constitutive NOS (cNOS), is an enzyme encoded by the NOS3 gene. Endothelial NOS generates nitric oxide in blood vessels and is involved with regulating vascular tone by inhibiting smooth muscle contraction and platelet aggregation. A constitutive calcium dependent NOS provides a basal release of NO. eNOS is associated with plasma membranes surrounding cells and the membranes of the Golgi apparatus within cells.

Used 1/500 dilution for WB and obtained a very clean and clear individual band at the expected MW. Regular Bis Tris Criterion Gel electrophoresis from BioRad was run with MES buffer. Immobilion transfer membrane purchased from Millipore.
