Antigen Affinity-purified
Immunogen
Mouse myeloma cell line NS0-derived recombinant mouse MSP R/Ron
Ser24-Arg960 (Ile372Leu)
Accession # CAA52754
Formulation
Lyophilized from a 0.2 μm filtered solution in PBS with Trehalose. *Small pack size (SP) is supplied either lyophilized or as a 0.2 µm filtered solution in PBS.
Endotoxin Level
<0.10 EU per 1 μg of the antibody by the LAL method.

Immunogen affinity purified

Applications

Recommended Concentration
Sample
Western Blot
0.1 µg/mL
Recombinant Mouse MSP R/Ron Fc Chimera (Catalog # 431-MS)
Neutralization
Measured by its ability to neutralize MSP-induced chemotaxis in mouse peritoneal macrophages [Wang, M-H. et al. (1994) J. Biol. Chem. 269:3436]. The Neutralization Dose (ND50) is typically 0.05-0.2 µg/mL in the presence of 50 ng/mL Recombinant Human MSP.

Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.

Chemotaxis Induced by MSP and Neutralization by Mouse MSP R/Ron Antibody. Recombinant Human MSP (Catalog # 352-MS) chemoattracts mouse peritoneal macrophages in a dose-dependent manner (orange line). The amount of cells that migrated through filter was measured by LeukoStat™ staining (Fisher Scientific). Chemotaxis elicited by Recombinant Human MSP (50 ng/mL) is neutralized (green line) by increasing concentrations of Mouse MSP R/Ron Antigen Affinity-purified Polyclonal Antibody (Catalog # AF431). The ND50 is typically 0.05-0.2 µg/mL.

Background: MSPR/Ron

Macrophage stimulating protein receptor (MSP R), encoded by the human RON and the mouse Stk, is one of a small family of receptor tyrosine kinases (RTKs) that also includes human Met (the receptor for hepatocyte growth factor) and chicken Sea. This family of receptors is synthesized as a single-chain precursor that is cleaved into a mature disulfide-linked heterodimer composed of an extracellular alpha chain and a membrane spanning beta chain with intrinsic tyrosine kinase activity. Mouse MSP R cDNA encodes a 1378 amino acid (aa) residue presursor protein with a 23 aa signal peptide, a 287 aa residue alpha chain (Ser 24 - Arg 310) and a 1068 aa residue transmembrane beta chain (Gly 311 - Thr 1378). Expression of MSP receptor is restricted to specific areas of the central and peripheral nervous systems, epithelial cells along the digestive tract, skin and lung, and in subpopulations of the mononuclear phagocyte lineage. Both the heterodimeric MSP and the free MSP beta chain have been shown to bind to MSP R. However, only the heterodimeric MSP binding can induce receptor dimerization and phosphorylation and cause biological activity.
