This DuoSet IC ELISA contains the basic components required for the development of sandwich ELISAs to measure total p21 / CIP1 / CDKN1A in cell lysates. An immobilized capture antibody specific for p21 / CIP1 / CDKN1A binds both phosphorylated and unphosphorylated p21 / CIP1 / CDKN1A. After washing away unbound material, a biotinylated detection antibody is used to detect both phosphorylated and unphosphorylated protein, utilizing a standard Streptavidin-HRP format.

p21/CIP1 is a Cyclin/Cyclin-dependent kinase (Cdk) inhibitor that blocks progression from G1 to S phase in the cell cycle. The expression of p21, a transcriptional target of p53, increases in cells where p53 is stabilized due to exposure to genotoxic agents.
