Principle of the Assay

The Proteome Profiler Rat Cytokine Array Kit, Panel A is a membrane-based sandwich immunoassay. Samples are mixed with a cocktail of biotinylated detection antibodies (Step 1) and then incubated with the array membrane which is spotted in duplicate with capture antibodies to specific target proteins (Step 2). Captured proteins are visualized using chemiluminescent detection reagents (Step 3). The signal produced is proportional to the amount of analyte bound.

Why Use an Antibody Array to Detect Multiple Cytokines?

Determining the expression of multiple cytokines in a single sample can be expensive, time consuming and can require specialized equipment. Performing multiple immunoprecipitations and Western blots requires time, labor, and reagents. The use of a multiplex antibody array to detect multiple cytokines in a single sample can be cost-effective and also save time and sample.

Scientific Data

The Rat Cytokine Array detects multiple analytes in cell culture supernates. NR8383 rat alveolar macrophage cells were either untreated or treated as below. Data shown are from a two minute exposure to X-ray film. A, untreated. B, Treated with 100 ng/mL LPS for 24 hours. C, Pre-treated with 100 nM Dexamethasone for 72 hours followed by treatment with 100 ng/mL LPS for 24 hours.

The Rat Cytokine Array detects multiple analytes in tissue lysates and serum. A rat was injected with 0.1 mg/kg LPS. After 24 hours, tissues were excised and prepared as described in the Sample Collection and Storage section. 400 µg of lysate was run on each array. Data shown are from a 7 minute exposure to X-ray film (Panels A-B). Tissues were excised from untreated rats and prepared as described in the Sample Collection and Storage section. 400 µg of lysate was run on each array. Data shown are from a 7 minute exposure to X-ray film (Panels C-E). Serum samples from 15 week old male rats were prepared as described in the Sample Collection and Storage section. 100 µL of serum was run on the array. Data shown are from a 5 minute exposure to X-ray film (Panel F).

The Rat Cytokine Array detects multiple analytes in cell culture supernates and lysates. (A. and B.) Supernates from NR8383 rat alveolar macrophage cell line untreated or treated with 50 ng/mL LPS for 24 hours. 500 μL of supernate was used for each array shown. (C. and D.) Lysates from NR8383 rat alveolar macrophage cell line untreated or treated with 50 ng/mL LPS for 24 hours. 200 μg of lysates was used for each array shown. Array images were collected and analyzed using the LI-COR Odyssey Infrared Imaging System.

The Rat Cytokine Array detects cytokines present in serum and tissue. Representative membranes and selective quantification are shown for rat serum (A., 150 μL), lysates of rat lung tissue (B., 400 μg), and lysates of rat liver tissue (C., 400 μg). Array images were collected and analyzed using the LI-COR Odyssey Infrared Imaging System.

  1. Will the Array identification number stamped on the Array membrane interfere with detection if it is not cut-off before the membrane is blocked?

    • The dye used for printing the Array identification number on the membranes will fluoresce and interfere with the LI-COR detection. It is critical that the number is cut off before beginning the experiment.

Performed well, great tool to screen for possible targets before making a more quantitative and more expensive assay. Spots can be easily evaluated using e.g., Bio-Rad ImageLab, and the semi-quantitative analysis can basically detect substantial differences between experimental groups.
